Seeking help replicating a Mendelian Randomization study on FKBP5 and reproductive outcomes (NEB, AFB, childlessness) Hi r/genetics, I'm an independent researcher in Japan and recently wrote a preprint proposing "putative Reproductive Inactivation Syndrome (pRIS)" — a framework suggesting that FKBP5 (a glucocorticoid receptor co-chaperone) acts as a key molecular transducer linking chronic stress (via HPA axis hyperactivity) to suppressed reproductive function (HPG axis). The paper uses: Two-sample MR (5-6 cis-eQTL instruments for FKBP5 expression from GTEx v8) Multi-variable MR (MVMR) adjusting for KISS1, CRH, and leptin Phenome-wide MR (PheWAS) across 45 phenotypes Some molecular docking for dietary polyphenols (e.g., curcumin) Key preliminary findings: Genetically predicted higher FKBP5 expression associated with fewer children ever born (NEB), earlier age at first birth (AFB), and higher odds of childlessness. MVMR suggested the effect is independent of kisspeptin, CRH, and leptin.…
